Hello everyone,
I have performed EMSA experiment with an RNA-binding protein and RNA. For both ssRNA and dsRNA, for higher concentration of protein, I observe a high intensity band at the dye front (lower than the free RNA band). The intensity of this band decreases with decrease in protein concentration. I have performed the experiments in a cold room with 0.5X TAE buffer and 8% gel.
Any idea why this is happening?
When coding qualitative and quantitative data, how would you link subject matter such as autonomy, being heard and listened to, independent life, decision-making, etc.? Every so often, someone presents work that is derivative of mine (e.g., Lightspeed Expanding Hyperspherical Universe or LEHU).
That particular hypothesis or postulate is impossible to defend. Richard Feynman proposed the hyperspherical topology in a lecture, but he couldn't defend it because of the problems it creates.
My theory - The Hypergeometrical Universe Theory (HU) solves those problems.
It emboldened many copycats who believed they could jump in the wagon and take part in the HU's model (for example, LEHU) and run.
By the way, I should clarify that LEHU is part of HU's model, not because I was the first to propose it, but because I am the only one who can defend it.
That creates flawed models where the flaws are obvious.
Here, I present a critique of one of these models. Feel free to disagree or agree in the comments.

It is my first time to deal with Label free proteomics data. The data was generated from Mayfly, it doesn't have annotated proteins. I used "uniprot_sprot.fasta" as a reference sequence, which has about 500k protein entry. The goal is to identify the protein and do differential analysis downstream. I used standard LFQ setting in maxquant and found only 85 entry in proteinGroups.txt file which is too small for the whole proteins in mayfly. When I run with a smaller set protein we have generated from Mayfly ( which partial protein) as a reference sequence, I got 330 entry in proteinGroups.txt. I expected a higher number of identified protein group when I use swiss sprot reference sequence. Any suggestion what might went wrong?

Hello, I have a question. I would appreciate you helping me. I want to do a Michael addition reaction between gelatin quantum dots with NH2 terminal groups and methyl acrylate. First, I synthesized gelatin quantum dots and determined the NH2 groups by ninhydrin assay; there was 228 mmol NH2 in 1mg of quantum dot; therefore, I reacted 0.5mg of GQD with 10.3 ml methylacrylate. Second, I did the Michael reaction in a free solvent and at room temperature with a 20% molar ratio of excess methyl acrylate to complete the reaction for a day. Notably, I hypothesize that gelatin quantum dots are like aniline. Thirdly, the analysis (ATR and NMR) was matched with methyl acrylate, and I guess that the Michael addition did not happen. Would you please help me?

What is a super vacuum? Is the earth in a vacuum? And what is dark energy?
It has not been proven until today and nature has always applied and proven exceptions and violations in the accepted theories many times in the past. That these were merely human formalisms and experimental artifacts and exploiting the limits of technology, and physical limits and laws are constantly being broken and bent in nature. Hereby we will attempt to show theoretically why and how there is and experimentally evidence in our universe of vacuum space, either in its theoretically idealized absolute form, thus free space or the partial vacuum that characterizes the vacuum of QED or QCD. And its zero-point energy and oscillations may actually be the greatest proof in nature for super energy.
It is possible without violating causation. that the apparent effect of "nothing" of vacuum space may be evidence for it
superluminocity and all this time it was hidden right in front of us. We are here trying to answer a fundamental question of physics, why the vacuum is basically space to us looks like nothing on the assumption that "nothing" exists in nature, and why a hypothetical superluminous vibration, a particle the size of Planck creates apparent nothingness in our spacetime. The novelty of the research here infers that free space is dark energy and that superluminous energy.

(1) Depends what is meant by ``super vacuum''. The words must, first, be defined, before questions can be asked. As it stands, it doesn't mean anything.
(2) To a good approximation the earth is moving around the Sun in a vacuum, i.e. its motion can be described by Newtonian mechanics, where the only bodies are the Earth and the Sun and the force between them is Newton's force of gravitation.
(3) Dark energy is the property of space and time that describes the fact that the Universe isn't, simply, expanding, but that this expansion is accelerating. To detect its effects it's necessary to measure the motion of bodies outside our galaxy.
To understand all this it's necessary to study classical mechanics-that leads to understanding the answer to the second question-and general relativity-in order to understand the answer to the third

Sometimes the people presenting certain untrustworthy offers of products or services are seemingly random people who, seemingly as mere citizens of the Internet who want to share their experiences of using various offers, presenting mainly or exclusively positive aspects of using certain presented products and services in reality are paid by the companies whose offers they present. Sometimes influencers and youtubers are given ownership of a specific advertised product for free as a form of gratification. In addition, artificial intelligence technology is increasingly being used to create advertising spots broadcast on social media. Individuals and companies using generative artificial intelligence technology, including applications based on AI technology available for free on the Internet to create advertising spots are taking advantage of legal loopholes, i.e. the lack of legal regulations that would normalize this sphere of the use of AI technology and limit the scale of misinformation, generation of fejknews, untrustworthy advertisements presenting various product and service offers using misleading content to the public that is inconsistent with facts, unverified using reliable, objective expert knowledge, scientific research conducted, etc. In addition, in AI-generated spots, animations and advertising videos, more and more often, instead of human influencers and youtubers, there are replacing them with a kind of avatars, digitally generated people who do not exist in reality. It happens that digitally generated images of real existing public figures of politicians, athletes, showbiz people, actors, singers, etc. are used in unreliably generated spots, animations and advertising videos, into whose mouths are put statements, texts, words that in reality they have never spoken. Recently, more and more often in the online social media, in which there are certain segments, generations of Internet users, citizens, there are many unreliable, taking advantage of the low level of knowledge in the field, offers of pseudo financial services, offers of supposedly super easy and highly profitable investments in cryptocurrencies, in miraculous investment strategies in Bitcoin requiring virtually no knowledge of finance, extra unique investment offers in precious metals, in contracts on selected securities, shares of dynamically growing startups basing their development on artificial intelligence technology, conducting innovative research projects with the aim of creating a miracle cure for cancer or other difficult-to-treat or incurable diseases. Public organizations and institutions representing the interests of consumer citizens, dealing with the issue of consumer protection and competition, investigating the problem of unreliable and misleading citizens presented in social media spots, animations, advertising videos, NGOs and socially active organizations are trying to warn citizens against such unreliable, fraudulent, false content ads. However, the main role in protecting citizens acting as consumers of information should be played by the technology companies running the aforementioned online social media. Leading online technology companies running popular social media sites are developing new technologies and are most equipped with modern ICT, Industry 4.0/5.0 technologies, and are therefore most predisposed to create reliably effective systems for verifying the content used in advertising campaigns run on their social media. This is because there is a lack of legal regulations in the legal normatives that would oblige the companies running social networks earning money from the advertisements posted to verify the content used in the advertisements, to check the issue of compliance of the content of the advertisements with the facts, with the generally applicable expert knowledge, with the results of scientific research conducted, and to make the said technology companies fully responsible for the content of the advertisements posted in their social media and for the financial, social, moral and other damages caused by the advertisements not properly verified. Besides, in addition to the necessary legal regulations, there should be a system of mandatory insurance fund financed by the said technology companies, from which compensation would be paid for all the negative effects caused by the broadcast on social media of fake news, misleading product and service offers, unreliable influencers, youtubers, advertising companies, etc. An additional solution that should be introduced is the possibility of legal enforcement of financial claims on the aforementioned insurance funds from unreliable influencers, youtubers, advertising companies, etc.

how many latitudes/longitudinal intersection lines are there?
The number of latitude and longitude lines can actually vary depending on how finely you want to divide the Earth's surface. Here are a few facts about these lines:
- There are 180 degrees of latitude, divided into 90 degrees north and 90 degrees south of the equator.
- There are 360 degrees of longitude, measured from the prime meridian (which runs through Greenwich, England) and going east or west.
- Together, these lines form a grid that can be used to identify any point on the Earth's surface using a set of coordinates.
- However, the number of intersection lines between latitude and longitude can be almost infinite, depending on how precisely you want to divide the grid. Typically, the grid is divided into degrees, minutes, and seconds - but it could be divided even further if needed.

1. I did the encapsulation of lemongrass essential oil microcapsule using chitosan and gum Arabic as wall material with different concentrations. I try to calculate the encapsulation efficiency using UV-vis spectrophotometer and get the absorbency. However, as I calculate using the EE(%) formula which is

the EE was low or even above 100. Can anyone enlighten me on which is the correct method to calculate the efficiency of this microcapsule using the absorbance result.

I am currently utilizing the R platform for my work. Leveraging free packages in R significantly aids in the successful completion of my tasks. I faced challenges while conducting Partial Least Squares (PLS) calculations on this particular platform. The two packages currently utilized are mixOmics and pls. However, these packages fail to meet the desired outcomes as indicated by previous research. The studies employ the PLS toolbox in Matlab for their analyses (https://doi.org/10.1016/j.jpba.2022.115037). I am facing a significant challenge due to the limited implementation budget, as the cost of the PLS toolbox exceeds $3,000. The financial implications associated with acquiring scientific research software in developing nations pose a significant obstacle to the advancement of scientific endeavors within these regions.
Does anyone possess any viable solutions to assist me in resolving the aforementioned issue? Are there any available packages or source script files for the PLS toolbox in R?

We have used CaFE Software to predict the binding free energy of our protein-ligand complexes from our molecular dynamics simulations.
Fortunately the Software has run successfully however don’t know the exact meaning of the physical thermodynamic parameters calculated: a) Complex total (kcal/mol), b) Receptor (kcal/mol), c) Ligand (kcal/mol) and d) Delta (kcal/mol).
We understand that the first value “Complex” can be interpreted as the binding free of the protein-ligand complex , However, we are bit confused about the last two terms “Ligand” (c) and Delta (d) what would be the difference between them? or which is the most important parameter to determine the binding free energy of the ligand to the receptor, is that (c) or (d) parameter?

The basic understanding of Islam shows and all Schools agree that measurability is essential condition for Riba. Unless one can measure something how does one know what is its Misl (equal). Riba is excess. In absence of measurement one can not know if there is excess. Absolute majority of our scholars do not know how to measure a free float. Prior to 1971/1914 we had no such problem and therefore other than our current understanding nothing other than Quran and Sunnah (including great work of our Fuqaha regarding its understanding and interpretation) is relevant in this case. The solution is simple consider fiat money to be what it is and apply Shariah on it according to its reality. Basic understanding of Riba shows that it can not be that it is not Ribawi. Accordingly it must be measurable as Shariah can never be wrong/irrelevant. You do not know how to measure ask those who know.
