If you want to generate graphics, there is PowerBI or Tableau, which are ideal for data visualization and dashboard generation (They have free licenses for students). Hello everyone,
I have performed EMSA experiment with an RNA-binding protein and RNA. For both ssRNA and dsRNA, for higher concentration of protein, I observe a high intensity band at the dye front (lower than the free RNA band). The intensity of this band decreases with decrease in protein concentration. I have performed the experiments in a cold room with 0.5X TAE buffer and 8% gel.
Any idea why this is happening?

Every so often, someone presents work that is derivative of mine (e.g., Lightspeed Expanding Hyperspherical Universe or LEHU).
That particular hypothesis or postulate is impossible to defend. Richard Feynman proposed the hyperspherical topology in a lecture, but he couldn't defend it because of the problems it creates.
My theory - The Hypergeometrical Universe Theory (HU) solves those problems.
It emboldened many copycats who believed they could jump in the wagon and take part in the HU's model (for example, LEHU) and run.
By the way, I should clarify that LEHU is part of HU's model, not because I was the first to propose it, but because I am the only one who can defend it.
That creates flawed models where the flaws are obvious.
Here, I present a critique of one of these models. Feel free to disagree or agree in the comments.

It is my first time to deal with Label free proteomics data. The data was generated from Mayfly, it doesn't have annotated proteins. I used "uniprot_sprot.fasta" as a reference sequence, which has about 500k protein entry. The goal is to identify the protein and do differential analysis downstream. I used standard LFQ setting in maxquant and found only 85 entry in proteinGroups.txt file which is too small for the whole proteins in mayfly. When I run with a smaller set protein we have generated from Mayfly ( which partial protein) as a reference sequence, I got 330 entry in proteinGroups.txt. I expected a higher number of identified protein group when I use swiss sprot reference sequence. Any suggestion what might went wrong?

I think the problem is mainly due to 'uniprot_sprot.fasta' having too few overlapped sequences with Mayfly (currently, there are only 3 proteins for Rhithrogena germanica and around 500 for Rhithrogena spp. in Uniprot. ). Therefore, only a few peptides are identified, and eventually, the FDR filtering kicks in early, cutting the list of identified peptides short. I would obtain the full protein sequence of Mayfly before conducting proteomics.

Hello, I have a question. I would appreciate you helping me. I want to do a Michael addition reaction between gelatin quantum dots with NH2 terminal groups and methyl acrylate. First, I synthesized gelatin quantum dots and determined the NH2 groups by ninhydrin assay; there was 228 mmol NH2 in 1mg of quantum dot; therefore, I reacted 0.5mg of GQD with 10.3 ml methylacrylate. Second, I did the Michael reaction in a free solvent and at room temperature with a 20% molar ratio of excess methyl acrylate to complete the reaction for a day. Notably, I hypothesize that gelatin quantum dots are like aniline. Thirdly, the analysis (ATR and NMR) was matched with methyl acrylate, and I guess that the Michael addition did not happen. Would you please help me?

Why is the protease inhibitor EDTA free? How does it make sure that metal ion dependent proteases are inhibited as well? They could chop down anything including capture antibodies.
Concerning "then Input there is no band at all" - did you check the protein transfer after Western Blot via ponceau to see if protein transfer was successful in general?
To optimize the protocol, I would prepare the beads first by blocking them via resuspension in albumin solution, wash once, then incubate with antibodies, then add to sample. otherwise a lot of unspecific binding might occur.

What is a super vacuum? Is the earth in a vacuum? And what is dark energy?
It has not been proven until today and nature has always applied and proven exceptions and violations in the accepted theories many times in the past. That these were merely human formalisms and experimental artifacts and exploiting the limits of technology, and physical limits and laws are constantly being broken and bent in nature. Hereby we will attempt to show theoretically why and how there is and experimentally evidence in our universe of vacuum space, either in its theoretically idealized absolute form, thus free space or the partial vacuum that characterizes the vacuum of QED or QCD. And its zero-point energy and oscillations may actually be the greatest proof in nature for super energy.
It is possible without violating causation. that the apparent effect of "nothing" of vacuum space may be evidence for it
superluminocity and all this time it was hidden right in front of us. We are here trying to answer a fundamental question of physics, why the vacuum is basically space to us looks like nothing on the assumption that "nothing" exists in nature, and why a hypothetical superluminous vibration, a particle the size of Planck creates apparent nothingness in our spacetime. The novelty of the research here infers that free space is dark energy and that superluminous energy.

(1) Depends what is meant by ``super vacuum''. The words must, first, be defined, before questions can be asked. As it stands, it doesn't mean anything.
(2) To a good approximation the earth is moving around the Sun in a vacuum, i.e. its motion can be described by Newtonian mechanics, where the only bodies are the Earth and the Sun and the force between them is Newton's force of gravitation.
(3) Dark energy is the property of space and time that describes the fact that the Universe isn't, simply, expanding, but that this expansion is accelerating. To detect its effects it's necessary to measure the motion of bodies outside our galaxy.
To understand all this it's necessary to study classical mechanics-that leads to understanding the answer to the second question-and general relativity-in order to understand the answer to the third

Individuals and companies using generative artificial intelligence technology, including applications based on AI technology available for free on the Internet to create advertising spots are taking advantage of legal loopholes, i.e. the lack of legal regulations that would normalize this sphere of the use of AI technology and limit the scale of misinformation, generation of fejknews, untrustworthy advertisements presenting various product and service offers using misleading content to the public that is inconsistent with facts, unverified using reliable, objective expert knowledge, scientific research conducted, etc. In addition, in AI-generated spots, animations and advertising videos, more and more often, instead of human influencers and youtubers, there are replacing them with a kind of avatars, digitally generated people who do not exist in reality. It happens that digitally generated images of real existing public figures of politicians, athletes, showbiz people, actors, singers, etc. are used in unreliably generated spots, animations and advertising videos, into whose mouths are put statements, texts, words that in reality they have never spoken. Recently, more and more often in the online social media, in which there are certain segments, generations of Internet users, citizens, there are many unreliable, taking advantage of the low level of knowledge in the field, offers of pseudo financial services, offers of supposedly super easy and highly profitable investments in cryptocurrencies, in miraculous investment strategies in Bitcoin requiring virtually no knowledge of finance, extra unique investment offers in precious metals, in contracts on selected securities, shares of dynamically growing startups basing their development on artificial intelligence technology, conducting innovative research projects with the aim of creating a miracle cure for cancer or other difficult-to-treat or incurable diseases. Public organizations and institutions representing the interests of consumer citizens, dealing with the issue of consumer protection and competition, investigating the problem of unreliable and misleading citizens presented in social media spots, animations, advertising videos, NGOs and socially active organizations are trying to warn citizens against such unreliable, fraudulent, false content ads. However, the main role in protecting citizens acting as consumers of information should be played by the technology companies running the aforementioned online social media. Leading online technology companies running popular social media sites are developing new technologies and are most equipped with modern ICT, Industry 4.0/5.0 technologies, and are therefore most predisposed to create reliably effective systems for verifying the content used in advertising campaigns run on their social media. This is because there is a lack of legal regulations in the legal normatives that would oblige the companies running social networks earning money from the advertisements posted to verify the content used in the advertisements, to check the issue of compliance of the content of the advertisements with the facts, with the generally applicable expert knowledge, with the results of scientific research conducted, and to make the said technology companies fully responsible for the content of the advertisements posted in their social media and for the financial, social, moral and other damages caused by the advertisements not properly verified. Besides, in addition to the necessary legal regulations, there should be a system of mandatory insurance fund financed by the said technology companies, from which compensation would be paid for all the negative effects caused by the broadcast on social media of fake news, misleading product and service offers, unreliable influencers, youtubers, advertising companies, etc. An additional solution that should be introduced is the possibility of legal enforcement of financial claims on the aforementioned insurance funds from unreliable influencers, youtubers, advertising companies, etc.

1/ - The answer to the main question is in the first sentence - companies that make money on advertising are materially interested in having as many ads as possible. Therefore, any verification of ads is not in their interest.
2/ - How do you imagine verifying the reliability of advertisements? If you do it really seriously, it may turn out that no ad will pass the reliability test. The essence of advertising is to persuade people to behave in a certain way, make decisions, make choices, make purchases, conclude contracts, etc. Advertising typically uses manipulation, at least by exposing the advantages and omitting the disadvantages of products, services, investment offers... etc. Expecting reliability from advertisements is at least naive, and making important decisions based on them without verification on your own is stupid.
3/ - AI is a threat because it raises the possibilities of manipulation to a higher level. But the possibilities of manipulation themselves have existed for a very long time - only the technical means of their implementation and propagation are changing. Without questioning the validity of attempts to legally regulate this phenomenon, I am afraid that without appropriate education these regulations will be ineffective. Ultimately, AI will certainly be a cheaper solution than corrupting the so-called influencers. There remains the issue of distinguishing real people from artificially generated personas. This problem has also existed for years, for example in the form of troll farms used in commercial activities, in political marketing, as well as for the manipulation of social moods and information and psychological operations, which we can beyond any doubt qualify as an element of hybrid warfare. AI provides new technical possibilities, but the phenomenon itself is not new.
4/ - The idea of compensation requires consideration of the scope of liability. Compensations may be counterproductive. People protected by the possibility of obtaining compensation may become less careful and prudent and make more risky decisions in hopes of compensating for possible negative consequences. The experience of numerous frauds and financial pyramids shows that people who are eager to look for easy profits are also eager to look outside for those who are guilty and responsible for their own mistakes. I believe that reinforcing such attitudes by shifting responsibility for wrong decisions to other entities is at least risky.
5/ - I see no chance for practical verification of the compliance of advertisements with scientific knowledge. The essence of advertising is a large dose of creativity aimed at giving the recipient specific impressions and emotions. Ultimately, the introduction of such regulations may end in questioning the image of a purple cow in the "Milka" advertisement, because there are no such cows, while the authors of the most perfidious advertisements will probably make every effort to maintain formal compliance with such regulations. When you order a service and you are not satisfied with its performance, you can easily verify whether the contractor simply did something wrong or wanted to cheat you. An honest contractor will try to solve the problem himself, while a fraudster will be so well prepared and legally protected that it may be unprofitable to get him to acknowledge the complaint. If someone intends to act dishonestly, they usually prepare well for it and take care to minimize legal risk. Therefore, I expect exactly the same with regard to possible regulations regarding advertising accuracy.
6/ - To sum up, I believe that trying to solve the problem using legal methods will not be effective. We must take into account great resistance from groups that earn a lot of money from this type of advertising, accusations of introducing censorship and limiting freedom of speech, as well as undesirable side effects. For example, attempts to combat disinformation and conspiracy theories by legal means may add media coverage and contribute to their popularization. It may also turn out that the introduced provisions will actually become the basis for limiting freedom of speech by, at least temporarily, blocking publications that are inconvenient for the authorities and do not necessarily have the nature of advertising content. Therefore, I believe that education related to this type of threats, developing critical thinking skills and verification of sources, as well as reliable economic and social education as an element of protection against manipulation and belief in easy profits or simple solutions to complex problems are much more important.

1. I did the encapsulation of lemongrass essential oil microcapsule using chitosan and gum Arabic as wall material with different concentrations. I try to calculate the encapsulation efficiency using UV-vis spectrophotometer and get the absorbency. However, as I calculate using the EE(%) formula which is

The terminology of FSPL somewhat misleading in my opinion. We do not loose energy because the there is some ‘loss’ along a path. The energy is lost, because some of it has traveled down a different path that no longer leads to our antenna. It is still there should we place more antenna in its way to receive it.
I have been careful in this post to only discuss things in freespace(i.e. a vacuum). when we start to add real world materials into the mix, things definitely diverge from this. House bricks definitely do increase the loss of signals that pass through them and this loss is very much dependent upon frequency, although not necessarily in a way that can be easily inserted into such a simple mathematical formula. However, when we start to think about propagation in environments that contain significant other materials, FSPL is probably not the way we want to estimate pathloss.

We have used CaFE Software to predict the binding free energy of our protein-ligand complexes from our molecular dynamics simulations.
Fortunately the Software has run successfully however don’t know the exact meaning of the physical thermodynamic parameters calculated: a) Complex total (kcal/mol), b) Receptor (kcal/mol), c) Ligand (kcal/mol) and d) Delta (kcal/mol).
We understand that the first value “Complex” can be interpreted as the binding free of the protein-ligand complex , However, we are bit confused about the last two terms “Ligand” (c) and Delta (d) what would be the difference between them? or which is the most important parameter to determine the binding free energy of the ligand to the receptor, is that (c) or (d) parameter?

I wanted to know a detailed protocol for the treatment of MDA-MB 231 cells with Free Fatty Acid (such as Palmitate, stearate, oleic acid, palmitoleic acid, etc.) . Thanks for the link. This seems a nice way to plot the poles given that the orientations are described in a .uxd format which are typically outputted by an x-ray diffraction experimental set-up. What i currently have is a plane formatted files that describes the orientations for each crystal in a single row for hkl crystal axis components. I would need to check the .uxd format first and find a way to port the plain format to this format in whatever way that seems viable. You could probably share your experience here.

desthiobiotin-labeled particles were enriched by streptavidin-coated magnetic beads in this study. Exosomes are eluted from beads by the application of a high concentration of biotin. This is a typical streptavidin/avidin-biotin affinity chromatography application. Desthiobiotin is replaced with biotin in seconds due to the high concentration of biotin. Desthiobiotin is a biotin analog that binds less tightly to biotin-binding proteins and is easily displaced by biotin. Both of the component has streptavidin binding affinity but a high concentration of biotin in this case represents a binding competition and replacement occurs between biotin and desthiobiotin.
In summary, high concentration and minor affinity differences are the keys to competitive binding herein.

Our beloved prophet (MPBUH) used to pray ' 'O Allah guide me to see the reality as it is and to form an opinion according to its reality'. We are confronted with freely floating fiat money. The basic understanding of Islam shows and all Schools agree that measurability is essential condition for Riba. Unless one can measure something how does one know what is its Misl (equal). Riba is excess. In absence of measurement one can not know if there is excess. Absolute majority of our scholars do not know how to measure a free float. Prior to 1971/1914 we had no such problem and therefore other than our current understanding nothing other than Quran and Sunnah (including great work of our Fuqaha regarding its understanding and interpretation) is relevant in this case. The solution is simple consider fiat money to be what it is and apply Shariah on it according to its reality. Basic understanding of Riba shows that it can not be that it is not Ribawi. Accordingly it must be measurable as Shariah can never be wrong/irrelevant. You do not know how to measure ask those who know.

Reality of gold and silver coins is their noble metal content. These buy what the contained quantity of gold/silver buys. Freely floating fiat money has no reality other than what it buys. Thus it can be measured only in terms of what it buys. If quantity of gold in gold coin is halved in Shariah it is equal to half of what it was. Similarly if the quantity of what free floats buy become half Shariah guides us to hold that it is Misl (equal) to half it was. Measuring gold coin by weight of its gold content is the best way to measure a gold coin. Similarly for silver coin best measure is the weight of it silver content. Reality of freely floating fiat money is not the paper what is recorded on it. Bank note and similar other representations are its record and not the reality. Thus if these are to be measured these must be measured in terms of their reality and that is what these buy.
Gold and silver coins also buy everything but are measured only in terms of their weight. Free floats buy all that is bought. Therefore meaning of what it buys may not be clear. Right interpretation is sum of all that is bought divided by total money used to buy. Full justification and explanation may please be seen in my work that is freely available and Insha Allah most relevant parts will be presented gradually.

I am starting with 220 emery paper to smooth the surface followed by 340, 400, 600, 800, 1200 with 200 rpm and 1500 and 2000 emery paper with 125 rpm followed by 3 micron but i am getting deep scratches on my sample.
If someone can suggest additional input how i can get scratch free sample for microscopy.

I would advise you to start with the emery paper of 600 till 2000 and do not use the 200, 300 and 400 ones.
You should follow this by metal polishing paste with rag wheel brush which is attached to a low speed dental motor to get a surface roughness of about 0.05 - 0.07 microns.
The emery paper especially the rough ones without the use of a polishing paste will cause surface scratches and high values of surface roughness.

As you know, in Abaqus, there are different types of connectors which, based on their free or constrained DOFs, we can define specific fasteners. The properties are defined in three main directions. Now my question is, how can we define a connector to work just in its force orientation ( it may be X, Y, or any other direction between these two main directions)?

The Translator connector type allows you to define a linear movement along a specified direction. Once you have defined the Translator connector, you can apply a force to it in any direction. The connector will then resist the force in the direction of the connector.
